I have created a data connection to a SharePoint list for a dropdown list field in a form, and have chosen “get choices from an external data source from the field properties, which is all working fine, but a requirement for this field is to also be able to ad unique vales, as well as being able to select from the dropdown. Is this possible?

You could use a Combo Box control in InfoPath to allow user to select an item from the dropdown OR type in a value themselves.

But if we use combo box,User can type the existing values and submit,how we will ensure that newly typed value is unique

The combo box doesn't have a way of knowing whether the data that has been filled out came via mouse selection in the combo box or whether the user typed the value in.

i.e. if my combo box has options 1, 2 and 3

I type in 2

Then I select the dropdown portion of the combo box, I'll see that the option for 2 is selected automatically.

Does that make sense?
