I am looking for a step by step guide to insert manager's email address from Active Directory.

I have used the GetUserProfileByName web service method to retrieve all other info, but cannot find a comprehensive guide to insert manager details.

I have found generic, filter again using manager account name, but no definitive guide.

 The issue is that manager email is not stored in Active Directory.

You could add it as a custom field.

Or you would need to be able to get the manager's username in AD and then requery for their user data secondarily.

Have you looked at Qdabra's AD Web Service?

Qdabra's AD Web Service will allow you to retrieve the alias of any user's manager, which you can then use to retrieve the manager's e-mail address. The web service also provides several other useful features and I recommend checking it out if you do a lot of work involving active directory.

OK i wrote something to give me the Direct Report Emails not to long ago. 

The managers e-mail is stored in Active Directory under the User Object of the Manager.  You can also use the displayName attribute + "@"domain.name to get the managers email unless of course it different. 

 Write a method using the DirectorySearcher Class.  You will do two ldap queries 1 on the direct report, and then query the manager for their email.
